1. What Is Glycine?

Glycine is a non-essential amino acid, meaning the body can make it from other molecules. It makes up roughly one-third of collagen protein and is abundant in skin, cartilage, bones, tendons and blood vessels. Beyond structure, glycine acts as:

  • An inhibitory neurotransmitter in the brainstem and spinal cord.
  • A precursor for glutathione, the body's master antioxidant.
  • A regulator of methionine metabolism via the methionine cycle.
  • A detoxification helper in the liver (conjugation of bile acids and xenobiotics).

Dietary sources include meat, fish, legumes, dairy and bone broth. Vegetarians and people on low-protein diets may have lower intakes.

2. Sleep and Fatigue Evidence

The strongest human evidence for glycine supplementation is in sleep. A series of Japanese studies found that 3g of glycine at bedtime:

  • Reduced the time to reach slow-wave sleep.
  • Improved subjective sleep quality and reduced morning fatigue.
  • Lowered core body temperature, a known sleep-promoting signal.

These effects are modest and most relevant for people with mild sleep complaints rather than diagnosed insomnia. Glycine is not a sedative and does not cause morning grogginess.

3. Collagen and Skin/Joints

Because glycine constitutes about one-third of collagen, glycine-rich collagen peptides are often studied for skin and joint outcomes. Pure glycine supplementation alone has less direct evidence for skin benefits than collagen peptides, but it supplies the raw material the body needs to make collagen. If your goal is skin or joint support, collagen peptides (which provide glycine plus proline and hydroxyproline) are usually the better-evidenced choice.

4. Longevity and Methionine Balance

High methionine intake (common in meat-heavy diets) may promote some ageing-related pathways in animals. Glycine is thought to buffer excess methionine by pushing it into useful metabolic pathways rather than harmful by-products. In rodents, adding glycine to a high-methionine diet partially mimics the benefits of methionine restriction, improving metabolic markers and possibly lifespan. Human translation is speculative, but the biology is plausible.

This is one reason glycine has become popular in longevity communities. However, no human trial has tested whether glycine supplementation extends lifespan.
