1. What Is NMNH?
NMNH (dihydronicotinamide mononucleotide) is a reduced form of nicotinamide mononucleotide. In the NAD+ biosynthetic pathway, NMN is normally converted into NAD+ by NMNAT enzymes. NMNH appears to bypass some rate-limiting steps more efficiently, producing larger and faster rises in intracellular NAD+ in preclinical models.
The compound first drew serious attention after a 2021 study in the FASEB Journal reported that NMNH was a "new and potent NAD+ precursor in mammalian cells and mice." Researchers found that NMNH raised NAD+ more effectively than NMN in several cell lines and increased liver NAD+ when given orally to mice.
2. The 2021 Breakthrough Study
Zapata-PΓ©rez and colleagues showed that:
- NMNH raised NAD+ levels faster than NMN in cultured human and mouse cells.
- Oral NMNH increased NAD+ in mouse liver and some other tissues.
- The effect was dose-dependent and could be blocked by inhibitors of the NAD+ salvage pathway.
This was enough to make NMNH the most-talked-about next-generation NAD+ precursor. However, the study did not measure lifespan, disease endpoints or long-term safety in animals, and no human data were included.
Bottom line: NMNH is a genuine scientific advance in NAD+ precursor chemistry, but it is not yet a proven human longevity intervention.
