Fisetin as a Senolytic: UK Evidence and Safety Guide

TL;DR: Fisetin is one of the most promising senolytic compounds in animal ageing research. It clears senescent cells, reduces inflammation and extends healthspan in mice. Human trials are ongoing; early data suggest safety, but we do not yet know whether fisetin clears zombie cells or extends healthspan in people. It should be considered experimental, not a proven longevity supplement.

1. What Are Senolytics?

Senescent โ€” "zombie" โ€” cells stop dividing but refuse to die. They secrete inflammatory molecules (the senescence-associated secretory phenotype, SASP) that contribute to tissue dysfunction, chronic inflammation and many age-related diseases. Senolytics are compounds that selectively clear these cells. Fisetin, quercetin + dasatinib (D+Q), and navitoclax are the best-known examples.

2. Fisetin Animal Evidence

Fisetin emerged as a leading senolytic after work from the Mayo Clinic and other labs:

  • Yousefzadeh et al., 2018: A EBioMedicine study screened flavonoids for senolytic activity and found fisetin to be among the most potent. In aged mice, fisetin improved tissue function, reduced senescence markers and extended median and maximum lifespan (doi:10.1016/j.ebiom.2018.09.015).
  • Healthspan improvements: treated mice showed better kidney, liver and lung function, reduced inflammation and preserved exercise capacity.
  • Late-life treatment: fisetin remained beneficial when started in already-old mice, a key hurdle for translation.

These results make fisetin one of the most exciting compounds in preclinical geroscience, but animal success frequently fails to translate to humans.

3. Human Trials: Mayo Clinic and Beyond

Several clinical trials are testing fisetin in humans:

  • NCT03430037 (Mayo Clinic): "Alleviation by Fisetin of Frailty, Inflammation, and Related Measures in Older Women." This phase II study tests intermittent high-dose fisetin in older women with frailty.
  • NCT04210986: A study of senolytic drugs, including fisetin, in osteoarthritis-related cartilage degeneration.
  • 2024 Mayo Clinic update: Preliminary reports suggest fisetin is safe and may reduce some markers of senescence and inflammation, but benefits appear heterogeneous โ€” not everyone responds.
โš ๏ธ Important: No published phase III trial has proven that fisetin clears senescent cells, prevents disease or extends life in humans.

4. Fisetin vs Quercetin + Dasatinib (D+Q)

The best-known human senolytic protocol is dasatinib + quercetin (D+Q), tested in small trials for diabetic kidney disease, idiopathic pulmonary fibrosis and osteoarthritis. Fisetin differs from D+Q in important ways:

  • Fisetin is a natural flavonoid found in strawberries, apples, grapes, onions and cucumbers. It has a better safety profile in animals than dasatinib, a chemotherapy drug.
  • D+Q has more human trial data, but dasatinib carries greater safety risk and is not available as a supplement.
  • Both are experimental. Neither is approved for anti-ageing use.

5. Dose and Protocols

The most discussed human protocol comes from Mayo Clinic trials: intermittent high-dose fisetin taken on two consecutive days per month, repeated monthly. For a 70 kg adult, this equates to roughly 1,000โ€“1,400 mg/day on dosing days, with no daily low-dose use.

This is not an established supplement regimen. The doses, frequency and long-term effects in healthy adults are unknown. UK food supplements typically sell 100โ€“500 mg/day capsules, which are lower than trial doses and have not been proven senolytic.

โš ๏ธ Do not self-prescribe: Taking fisetin at research-level intermittent doses without medical supervision is experimental. Speak to a clinician, especially if you take blood thinners, chemotherapy or other medications.

6. Safety and Interactions

Fisetin is generally recognised as safe as a food component, but high-dose supplement use is experimental. Considerations:

  • Blood thinners: fisetin may have antiplatelet effects; use caution with warfarin, apixaban, rivaroxaban and aspirin.
  • Chemotherapy and radiation: senolytics may theoretically alter cancer risk or treatment response; do not use without oncology advice.
  • Pregnancy and breastfeeding: no safety data; avoid.
  • Surgery: stop high-dose fisetin before surgery due to bleeding risk.

7. Buying Fisetin in the UK

Fisetin is sold as a food supplement in the UK. If you and a clinician decide to try it:

  • Look for high-purity extracts: 98%+ fisetin, often from Sophora japonica (Japanese pagoda tree).
  • Check dose: many capsules contain 100โ€“500 mg; be aware this is below trial intermittent doses.
  • Third-party testing: certificates of analysis for purity and heavy metals.
  • UK seller: easier returns and regulatory recourse.

Look for third-party-tested UK products from sellers that publish certificates of analysis and have a UK-registered business address.
