1. What Is Ca-AKG?
Alpha-ketoglutarate (AKG) is a five-carbon intermediary metabolite in the Krebs cycle. It sits at the crossroads of energy production, amino-acid metabolism and epigenetic clocks regulation. Calcium alpha-ketoglutarate (Ca-AKG) is the calcium salt, which is less acidic, better tolerated in the gut and supplies a small amount of elemental calcium.
AKG declines with age in humans and other mammals. It also acts as a substrate for dioxygenase enzymes involved in DNA demethylation and collagen peptides guide hydroxylation, giving it roles in both energy metabolism and tissue maintenance.
2. The 2020 Mouse Lifespan Study
The most cited Ca-AKG longevity paper comes from Buck Institute researchers and was published in Cell Metabolism in 2020. Middle-aged female mice given AKG in their drinking water showed:
- Longer median lifespan. The treated group lived roughly 12% longer than controls.
- Compressed morbidity. AKG mice spent less of their life in a frail state, suggesting improved healthspan.
- Reduced systemic inflammation. Lower levels of inflammatory markers, including IL-6.
- Better bone and gut barrier markers. Suggesting multi-system effects.
Importantly, the benefit was seen when treatment started at middle age, not when it began very late in life. Males showed some but not all of the same effects.

Human Ca-AKG trials are smaller and newer than the mouse work. As of 2026, completed studies focus on biomarkers rather than lifespan.
- DNA methylation clocks. A 2021 open-label study of a Ca-AKG + vitamin formulation (Rejuvant®) reported an average reduction in biological age of roughly 8 years over 7 months, measured by a DNA methylation clock (Demidenko et al., 2021). This was not placebo-controlled, so regression to the mean and placebo effects cannot be ruled out.
- Bone turnover. A 2016 trial in postmenopausal women with osteopenia found that Ca-AKG reduced C-terminal cross-linking telopeptide of type I collagen (CTX), a marker of bone resorption, over six months.
- Post-surgery trials. Ongoing trials such as AEGIS and ABLE are testing whether AKG can improve recovery and biological-age markers in older adults undergoing or recovering from surgery.
4. Proposed Mechanisms
Why might AKG affect ageing? Several non-exclusive mechanisms have been proposed:
- Krebs-cycle fuel. AKG helps mitochondria produce ATP and may compensate for age-related metabolic slowing.
- Epigenetic regulation. AKG is a cofactor for TET and JmjC demethylases that regulate DNA and histone methylation.
- Prolyl hydroxylase substrate. Required for collagen formation and hydroxylation reactions.
- Inflammation modulation. Animal studies suggest reduced IL-6 and NF-κB signalling.
- mTOR and autophagy. Some preclinical work hints at nutrient-sensing effects, though human confirmation is lacking.
5. Dose and Supplement Forms
Most longevity supplements provide 500 mg to 1,000 mg of Ca-AKG per day, split into one or two servings. This range overlaps with amounts used in human biomarker studies. There is no established RDA or upper safe limit from regulators. This is research data, not a recommendation.

Ca-AKG has a short but reassuring safety record in human trials. The most common side effects are mild and gastrointestinal:
